Understanding axonal growth and guidance is essential both for the comprehension of how neural circuits are built up, and also for the identification of mechanisms responsible for the lack of axonal regeneration in the adult CNS. The characterization of the biochemical cascades required for axonal growth may lead to the development of bioactive compounds and therapeutical approaches to stimulate the re-growth of damaged connections. The chemoattractive molecule Netrin 1 is essential for the formation of many axonal pathways in the brain, and acts through specific receptors that mediate chemoattraction (DCC) or chemorepulsion (UNC5H). From a conceptual point of view, axonal growth and guidance may require an exquisite regulation of the exocytotic machinery that controls plasma membrane addition. This project proposal aims to study the link between the exocytotic machinery at the growth cone and Netrin 1-mediated axonal growth and guidance. We have demonstrated that the Netrin 1 receptor DCC associates in vitro and in vivo with the SNARE proteins Syntaxin 1A and 1B, and that loss of Syntaxin function in neuronal cultures abolishes Netrin 1-induced chemoattraction. We aim to elucidate this new signalling mechanism at the molecular and cellular levels, and to determine the possible participation of other proteins which control Syntaxin functions. The identification of this novel signalling system may open up new strategies for axonal repair and regeneration
